Application of Chlorophyll Analyzer in Processing Technology of Toona Leaf
In the process of extraction, processing, storage and use of chlorophyll from Toona sinensis leaves, it should be carried out under the conditions of protection from light, low temperature and neutrality as much as possible to prevent adverse effects of various ions on it. At the same time, for citron leaves, the chlorophyll meter assay after the extraction is also affected by some of the above factors.
The study on the extraction conditions of chlorophyll of Toona sinensis leaves showed that the optimum extraction process of chlorophyll from Toona sinensis leaves was ethanol: extraction temperature 60 °C, extraction time 4.0 h. Under optimal conditions, the chlorophyll absorbance value of Toona sinensis leaves was 1.609. The study on the stability of chlorophyll of Toona sinensis leaves showed that the impact of light on the stability of chlorophyll of Toona sinensis leaves and the determination of chlorophyll meter, when the illumination time was up to 4h, the chlorophyll absorbance value dropped sharply.
With the increase of the exposure time of chlorophyll under natural light conditions, the absorbance of chlorophyll extract continues to decrease. The test of chlorophyll meter can also indicate that the value of chlorophyll absorbance drops to 0.201 when light is irradiated for 4 hours. The absorbance value of chlorophyll before 85 °C decreased obviously, but the absorbance value decreased slightly after 85 °C incubation, that is, the chlorophyll of Toona sinensis leaves had certain high temperature resistance.
Chlorophyll measurement showed that the absorbance of chlorophyll decreased significantly before 85°C, but the absorbance decreased slightly after 85°C, that is, the chlorophyll of Toona sinensis leaves had a certain high temperature resistance; the stability of chlorophyll in pH value of Toona sinensis leaves was detected by pH. The effect is larger. With the increase of pH, the absorbance value of chlorophyll of Toona sinensis leaves is also increasing. When the pH is more than 7, the absorbance of the chlorophyll decreases obviously. The chlorophyll of Toona sinensis leaves has better resistance to reduction.
